The Buffalo Bills selected Hamlin with the 212th overall pick in the sixth round of the 2021 National Football League Draft. He signed a four-year rookie contract with Buffalo on May 21, 2021.

Hamlin was one of the top cornerbacks after graduating from Central Catholic. He was a four-star recruit for prestigious colleges that play football, like Pittsburgh, Penn State, Ohio State, and Temple. He decided to enrol at Pitt in order to play collegiate football with the Panthers. He played in three games prior to getting hurt and redshirting as a freshman.

Hamlin passed out after colliding with wide receiver Tee Higgins on January 2, 2023, during a Monday Night Football game against the Cincinnati Bengals during Week 17 of the National Football League. Hamlin was smacked in the chest by Higgins’ leading right shoulder.

Hamlin then dragged Higgins down by entangling his arms around his shoulders and helmet. Despite highlighting the striking’s shocking severity, sources described the tackle as routine. Shortly after the auction, Hamlin stood up before bending over to his back.

Prior to the arrival of an ambulance, medical staff appeared to have performed CPR on Hamlin on the field for around 10 minutes.
