On January 19, 1980, Thomas Luke Macfarlane was born in London, Ontario. His mother Penny works as a mental health nurse in a London hospital, and his father Thomas served as the director of student health services at the University of Western Ontario. Along with her older sister Rebecca and identical twin sister Ruth, Macfarlane attended London Central Secondary School. Macfarlane was the lead singer and songwriter for the band Fellow Nameless, which was founded in his eighth grade along with some of his classmates at Lester B. Pearson School for the Arts under the name Slipnaught, a name they arbitrarily picked out of a dictionary because they did not have a name for the band when it came time to perform on stage.
From November 14 to December 21, 2003, Macfarlane was one of four leads in Juvenilia at Playwrights Horizons Theater. In May 2004, Luke played the lead in the American premiere of the play Where Do We Live, which was staged at the Vineyard Theatre. The 2005 GLAAD Media Awards recognized the production as Outstanding New York Theatre: Broadway and Off-Broadway. In Summer 2006, Luke co-starred with Jill Clayburgh and Hamish Linklater in the off-Broadway production of The Busy World is Hushed at Playwrights Horizons. From February 7 to 11, 2007, Luke reprised his role as Thomas at the Skirball Cultural Center in Los Angeles.
