Thomas Luke Macfarlane was born in London, Ontario on January 19, 1980. Thomas, his father, was the Director of Student Health Services at the University of Western Ontario, and Penny, his mother, is a mental health nurse at a London hospital. Macfarlane went to London Central Secondary School with her twin sister Ruth and her older sister Rebecca. Macfarlane attended the Lester B. Pearson School for the Arts before studying drama at Juilliard in New York City.

From November 14 to December 21, 2003, Macfarlane was one of four leads in Juvenilia at Playwrights Horizons Theater. In May 2004, Luke played the lead in the American premiere of the play Where Do We Live, which was staged at the Vineyard Theatre. The 2005 GLAAD Media Awards recognized the production as Outstanding New York Theatre: Broadway and Off-Broadway. In Summer 2006, Luke co-starred with Jill Clayburgh and Hamish Linklater in the off-Broadway production of The Busy World is Hushed at Playwrights Horizons. From February 7 to 11, 2007, Luke reprised his role as Thomas at the Skirball Cultural Center in Los Angeles.

Luke Macfarlane attended London Central Secondary School alongside his twin sister Ruth Macfarlane and older sister Rebecca Macfarlane. Luke later graduated from the Lester B. Pearson School for the Arts, after which he studied drama at the Juilliard School of New York. Luke was naturalized as an American citizen on June 12, 2018.
