Karl-Anthony played basketball for the Kentucky Wildcats in college. Karl-Anthony was chosen for the Dominican Republic national team at the age of 16. He was chosen by the Minnesota Timberwolves with the first overall pick in the 2015 NBA Draft, and for the 2015–16 season, he was named NBA Rookie of the Year.

Karl-Anthony has received three All-Star game selections. In 2021–2022, Towns won the NBA Three-Point Contest. He has been the Dominican Republic’s ambassador overseas since 2012. He made his debut overseas at the 2012 Centrobasket when he was 16 years old.
